Phospho-MLKL (S345) Recombinant Rabbit Monoclonal Antibody [JM92-37] (ET1705-51)

  • Recombinant

Applications

  • IHC-P

REACTIVITY

  • Mouse

Immunohistochemical analysis of paraffin-embedded mouse spleen tissue using anti-MLKL (phospho S345) antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0-8.4) for 20 minutes. The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (ET1705-51, 1/50) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

SEQUENCE SIMILARITIES

Belongs to the protein kinase superfamily.

TISSUE SPECIFICITY

Highly expressed in thymus, colon, intestine, liver, spleen and lung. Expressed at much lower level in skeletal muscle, heart and kidney. Not detected in brain.

POST-TRANSLATIONAL MODIFICATION

Phosphorylation by RIPK3 induces a conformational switch that is required for necroptosis. It also induces homotrimerization and localization to the plasma membrane.

SUBCELLULAR LOCATION

Cytoplasm, Membrane.

FUNCTION

Pseudokinase that plays a key role in TNF-induced necroptosis, a programmed cell death process. Activated following phosphorylation by RIPK3, leading to homotrimerization, localization to the plasma membrane and execution of programmed necrosis characterized by calcium influx and plasma membrane damage. Does not have protein kinase activity.
