HUABIO Quality Standards — 5+1 Antibody Validation Strategy

At HUABIO, we refuse to create antibodies that contribute to our industry's problem of irreproducible results and instead are determined to deliver heavily tested and consistent products directly to your lab. We're always challenging ourselves to be more efficient and accurate while routinely evolving our products for researchers all over the world.

Building upon the principles outlined in 'A proposal for validation of antibodies' (Nature Methods, 2016), we have developed a '5+1' strategy aimed at verifying the functionality, specificity, and sensitivity of each antibody we produce. This strategy is upheld by five fundamental pillars, all working in unison to achieve our overarching objective: helping our customers to consistently attain reproducible results, from the first purchase and every reorder after.

Genetics

Knockout or knockdown the target gene by using CRISPR/Cas9 or RNAi technology in cells or tissues, and validate the antibody specificity by measuring the absence or reduction of the antibody signal in these cells or tissues.

Biological Properties and Orthogonal Data

Qualitative and quantitative verification based on biological properties, or use antibody-independent assays to detect correlations between antibodies and biological properties or non-antibody quantification.

Multiple Antibodies

Validate by using two (or more) independent antibodies that can recognize different epitopes of the target protein.

Heterologous Expression

Antibody signal is evaluated in cell lines following heterologous expression of native (or mutated) target protein.

Complementary Validation

Antibody specificity may be validated using complementation assays. Including competitive ELISA, peptide blocking, or protein arrays.

Recombinant Production

Provides researchers with advantages which include superior lot-to-lot consistency, continuous supply, and amenability to antibody engineering.


Stability

We examined the stability of our antibodies at different storage temperatures for varying lengths of time to verify the stability and quality of antibodies during transportation and storage. We have demonstrated that with a subset of our antibodies, room temperature does not impair their activity for up to three months.

Specificity

Knockout (KO) validation is used to confirm antibody specificity by testing the antibody of interest in a KO cell line or tissue that does not express the target protein. No signal is observed when a specific antibody is tested in a KO cell line while giving the specific target protein signal in the wild-type (WT) cell line.

Lot-to-Lot Consistency

A fundamental advantage of recombinant antibodies is their superior lot-to-lot consistency.
