REACTIVITY

Western blot analysis of Cytokeratin-7 on different lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody (EM0702, 1/500) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1:200,000 dilution was used for 1 hour at room temperature.<br />

EM0702 was shown to specifically react with Cytokeratin-7 in wild-type Hela cells. No band was observed when Cytokeratin-7 knockout samples were tested. Wild-type and Cytokeratin-7 knockout samples were subjected to SDS-PAGE. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM in TBST for 1 hour at room temperature. The primary antibody (EM0702, 1/1,000) and Loading control antibody(Rabbit anti-Vinculin, ET1705-94, 1/5,000) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Mouse IgG-HRP Secondary Antibody (HA1006) at 1:20,000 dilution was used for 1 hour at room temperature.
  • Immunohistochemical analysis of paraffin-embedded human lung bronchus tissue using anti-Cytokeratin-7 antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0-8.4) for 20 minutes.The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (EM0702, 1/200) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

TISSUE SPECIFICITY

Expressed in cultured epidermal, bronchial and mesothelial cells but absent in colon, ectocervix and liver. Observed throughout the glandular cells in the junction between stomach and esophagus but is absent in the esophagus.

FUNCTION

Keratin-7 is a member of the keratin gene family. The type II cytokeratins consist of basic or neutral proteins which are arranged in pairs of heterotypic keratin chains coexpressed during differentiation of simple and stratified epithelial tissues. Keratin-7 is found in simple glandular epithelia, and in transitional epithelium. Epithelial cells of the lung and breast both contain keratin-7, but some other glandular epithelia, such as those of the colon and prostate, do not. Because the keratin-7 antigen is found in both healthy and neoplastic cells, antibodies to CK7 can be used in immunohistochemistry to distinguish ovarian and transitional cell carcinomas from colonic and prostate cancers, respectively.
