Function
The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. Members of the PTP family share a highly conserved catalytic motif, which is essential for the catalytic activity. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. Epidermal growth factor receptor and the adaptor protein Shc were reported to be substrates of this PTP, which suggested the roles in growth factor mediated cell signaling. Multiple alternatively spliced transcript variants encoding different isoforms have been found. Two highly related but distinctly processed pseudogenes that localize to chromosomes 1 and 13, respectively, have been reported.

Western blot analysis of TCPTP on different lysates with Rabbit anti-TCPTP antibody (HA720074) at 1/5,000 dilution.
Lane 1: K-562 (Human chronic myelogenous leukemia cell) cell lysate
Lane 2: NIH/3T3 (Mouse fibroblast) cell lysate
Lysates/proteins at 10 µg/Lane.
Exposure time: 20 seconds; ECL: K1801
Blocking: 5% NFDM/TBST, 1 hour at room temperature
Primary antibody: HA720074, 1/5,000 in primary antibody dilution buffer 5% NFDM/TBST, overnight at 4 ℃
Secondary antibody: Goat anti-Rabbit IgG-HRP (HA1001), 1/50,000 in 5% NFDM/TBST, 1 hour at room temperature
Predicted band size: 48.5 kDa
Observed band size: 45, 48 kDa -
Flow cytometric analysis of Daudi cells labeling TCPTP.
Cells were fixed and permeabilized. Then stained with the primary antibody (HA720074, 1ug/ml) (red) compared with Rabbit IgG Isotype Control (green). After incubation of the primary antibody at +4℃ for an hour, the cells were stained with a iFluor™ 488 conjugate-Goat anti-Rabbit IgG Secondary antibody (HA1121) at 1/1,000 dilution for 30 minutes at +4℃. Unlabelled sample was used as a control (cells without incubation with primary antibody; black).
