Function

In eukaryotes, the phosphorylation and dephosphorylation of proteins on serine and threonine residues is an essential means of regulating a broad range of cellular functions including division, homeostasis and apoptosis. A group of proteins that are intimately involved in this process are the protein phosphatases. In general, the protein phosphatase (PP) holoenzyme is a trimeric complex composed of a regulatory subunit, a variable subunit and a catalytic subunit. Four major families of protein phosphatase catalytic subunit have been identified, designated PP1, PP2A, PP2B and PP2C. An additional protein phosphatase catalytic subunit, PPX, is a putative member of a novel PP family. PP6, also known as PPP6C, is a 305 amino acid cytoplasmic protein that belongs to the PPP phosphatase family. Ubiquitously expressed, PP6 is component of a signaling pathway regulating cell cycle progression in response to IL-2 receptor stimulation and is involved in suppressing inflammatory responses by specifically down-regulating TR4.

  • Western blot analysis of PPP6C on different lysates. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (<a href="/products/HA720056" style="font-weight: bold;text-decoration: underline;">HA720056</a>, 1/500) was used in 5% NFDM/TBST at room temperature for 2 hours.   • Immunohistochemical analysis of paraffin-embedded human kidney tissue using anti-PPP6C antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes.The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH<sub>2</sub>O and PBS, and then probed with the primary antibody (<a href="/products/HA720056" style="font-weight: bold;text-decoration: underline;">HA720056</a>, 1/50) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

  • Immunocytochemistry analysis of SiHa cells labeling PPP6C with Rabbit anti-PPP6C antibody (<a href="/products/HA720056" style="font-weight: bold;text-decoration: underline;">HA720056</a>) at 1/50 dilution.<br /><br />Cells were fixed in 4% paraformaldehyde for 10 minutes at 37 ℃, permeabilized with 0.05% Triton X-100 in PBS for 20 minutes, and then blocked with 2% negative goat serum for 30 minutes at room temperature. Cells were then incubated with Rabbit anti-PPP6C antibody (<a href="/products/HA720056" style="font-weight: bold;text-decoration: underline;">HA720056</a>) at 1/50 dilution in 2% negative goat serum overnight at 4 ℃. Goat Anti-Rabbit IgG H&L (Alexa Fluor&reg; 488) was used as the secondary antibody at 1/1,000 dilution. Nuclear DNA was labelled in blue with DAPI.<br /><br />Beta tubulin (<a href="/products/M1305-2" style="font-weight: bold;text-decoration: underline;">M1305-2</a>, red) was stained at 1/200 dilution overnight at +4℃. Goat Anti-Mouse IgG H&L (Alexa Fluor&reg; 647) were used as the secondary antibody at 1/1,000 dilution.

Cells were then incubated with Rabbit anti-PPP6C antibody (HA720056) at 1/50 dilution in 2% negative goat serum overnight at 4 ℃. Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) was used as the secondary antibody at 1/1,000 dilution. Nuclear DNA was labelled in blue with DAPI.

    Beta tubulin (M1305-2, red) was stained at 1/200 dilution overnight at +4℃. Goat Anti-Mouse IgG H&L (Alexa Fluor® 647) were used as the secondary antibody at 1/1,000 dilution.
