HOXA9, also known HOX1G, which belongs to the Abd-B homeobox family and transiently interacts with PRMT5 in TNF-alpha stimulated endothelial cells. HOXA9 is methylated on Arg-140 by PRMT5and methylation is critical for E-selectin induction. HOXA9 as a sequence-specific transcription factor which is part of a developmental regulatory system that provides cells with specific positional identities on the anterior-posterior axis. HOXA9 is required for induction of E-selectin and VCAM-1, on the endothelial cells surface at sites of inflammation. HOXA9 is 30 kDa, but methylation of HOXA9 is about 35-40 kDa.

Immunohistochemical analysis of paraffin-embedded mouse kidney tissue using anti-HOXA9 antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) (high pressure) for 2 minutes. The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HA500077, 1/400) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
