5 tips for IHC-P

Immunohistochemistry-Paraffin (IHC-P) is a widely used technique that can provide valuable insight into the distribution and expression of antigens within tissue samples. By utilizing specific antibodies and a series of well-established protocols, IHC-P enables researchers to visualize the localization and presence of proteins, hormones, and other molecular markers. Despite its many benefits, IHC-P can also present challenges, such as optimizing staining conditions and avoiding non-specific binding, that must be taken into consideration for obtaining reliable results. 

Here are a few tips for a successful experiment:

  • Fix it to fix it: Proper fixation is key to getting great IHC-P results. So make sure to choose the right fixative for your tissue and antigen of interest and stick to the recommended fixation time.  Read more about fixation HERE.


  • Don't hide the antigens: Antigen retrieval can be a lifesaver when it comes to unmasking antigens that may have become inaccessible during fixation. So don't be afraid to heat things up a bit or try a different method to get the best results. Read more about antigen retrieval HERE.


  • Choose wisely: Selecting the right primary antibody is like choosing the right partner. You want to make sure it's specific and validated for the tissue type and species you are studying. And always check its credentials before jumping in!


  • Block it out: Blocking non-specific binding with serum from the species in which the secondary antibody was generated is like putting up a wall to keep the bad guys out. Make sure to do it properly for the best results. Read more about blocking HERE.


  • Experiment and optimize: Optimizing staining conditions is like cooking the perfect dish. Try different incubation times, temperatures, and concentrations of primary and secondary antibodies until you find the sweet spot for your IHC-P.
