Function
Chemokines are members of a superfamily of inducible, secreted, pro-inflammatory cytokines. Members of the chemokine family exhibit 20% to 50% homology in their predicted amino acid sequences and are divided into four subfamilies: C-C, C-X-C, C and C-X3-C. In the C-X-C or α subfamily, the first two of four cysteine motifs are separated by another amino acid residue. In the second subfamily, designated C-C or β, the first cysteines are adjacent. C subfamily members, also designated γ chemokines, lack the first and third cysteine residues of the conserved motif. In the C-X3-C, or δ subfamily, members have three amino acids between the two cysteines. The C-X-C chemokine subfamily includes IL-8, GROα/β/γ (and the murine homologs KC, MIP-2α and MIP-2β), platelet basic protein, ENA-78, GCP-2, PF4, IP-10 (and its murine homolog, CRG) and MIG.

Tissue Specificity
Mainly secreted by monocytes, endothelial cells as well as fibroblasts. Expressed by epithelial cells in thymus. Microglial cells produce CXCL10 in response to viral stimulation.
Post-translational Modification
Several proteases can mediate post-secretion cleavages. DPP4 cleaves CXCL10 on its N-terminal 2 amino acids leading to an antagonist form of CXCL10. This dominant negative form is capable of binding CXCR3 but does not induce signaling. MMP9 cleaves 9 amino acids instead.

Western blot analysis of CXCL10/IP10 on different lysates with Rabbit anti-CXCL10/IP10 antibody (ET1704-27) at 1/1,000 dilution.
Lane 1: THP-1 cell lysate
Lane 2: THP-1 treated with 100ng/mL IFN gamma for 24 hours then 300ng/mL BFA for 20 hours cell lysate
Lane 3: THP-1 cell lysate
Lane 4: THP-1 treated with 200ng/mL IFN gamma and 50ng/mL LPS for 24 hours cell lysate

4-20% SDS-PAGE gel.
Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (ET1704-27) at 1/1,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1/50,000 dilution was used for 1 hour at room temperature. -

Immunocytochemistry analysis of THP-1 cells treated with 200ng/mL IFN gamma then treated with 50ng/mL LPS for 24 hours labeling CXCL10/IP10 with Rabbit anti-CXCL10/IP10 antibody (ET1704-27) at 1/100 dilution.
Cells were fixed in 4% paraformaldehyde for 20 minutes at room temperature, permeabilized with 0.1% Triton X-100 in PBS for 5 minutes at room temperature, then blocked with 1% BSA in 10% negative goat serum for 1 hour at room temperature. Cells were then incubated with Rabbit anti-CXCL10/IP10 antibody (ET1704-27) at 1/100 dilution in 1% BSA in PBST overnight at 4 ℃. Goat Anti-Rabbit IgG H&L (iFluor™ 488, HA1121) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. Nuclear DNA was labelled in blue with DAPI. -
Immunohistochemical analysis of paraffin-embedded human tonsil tissue with Rabbit anti-CXCL10/IP10 antibody (ET1704-27) at 1/1,000 dilution.
The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (ET1704-27) at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX. -
