CD14 is a glycophosphatidylinositol-anchored protein expressed on the surfaces of monocytes and plymorphonuclear leukocytes. It cooperates with MD-2 and TLR4 to mediate the innate immune response to bacterial lipopolysaccharide (LPS). CD14 acts via MyD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response and up-regulates cell surface molecules, including adhesion molecules.

Tissue Specificity
Detected on macrophages (at protein level). Expressed strongly on the surface of monocytes and weakly on the surface of granulocytes; also expressed by most tissue macrophages.
Post-translational Modification
N- and O- glycosylated. O-glycosylated with a core 1 or possibly core 8 glycan.
Western blot analysis on K562 cell lysates using anti-CD14 rabbit polyclonal antibody.
ICC staining CD14 in SW480 cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.
