Function
RNA-binding protein Musashi homolog 1 also known as Musashi-1 is a protein that in humans is encoded by the MSI1 gene. This gene encodes a protein containing two conserved tandem RNA recognition motifs and functions as an RNA binding protein that is involved in post-transcriptional gene editing. It is a stem cell marker that controls the balance between self-renewal and terminal differentiation. Over expression of this gene is associated with the grade of the malignancy and proliferative activity in gliomas and melanomas. An increased expression of MSI1 protein is observed in endometriosis and endometrial carcinoma siRNA-mediated inhibition of MSI expression in endometrial carcinoma cells induces apoptosis and inhibits cell proliferation by affecting the Notch signaling pathway. MSI1 is highly expressed in neural progenitor cells and is required for normal development of the brain. A mutation in these gene is responsible for autosomal recessive primary microcephaly. MSI1 also interacts with the Zika virus genome and may explain why these cells are highly susceptible to Zika virus infection.

Western blot analysis of Musashi 1 on different lysates with Rabbit anti-Musashi 1 antibody (ET1606-51) at 1/2,000 dilution.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (ET1606-51) at 1/2,000 dilution was used in 5% NFDM/TBST at 4℃ overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1/50,000 dilution was used for 1 hour at room temperature. -
Immunohistochemical analysis of paraffin-embedded mouse cerebellum tissue with Rabbit anti-Musashi 1 antibody (ET1606-51) at 1/200 dilution.
The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (ET1606-51) at 1/200 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX. -

Flow cytometric analysis of Musashi 1 was done on Hela cells. The cells were fixed, permeabilized and stained with the primary antibody (ET1606-51, 1/50) (red). After incubation of the primary antibody at room temperature for an hour, the cells were stained with a Alexa Fluor 488-conjugated Goat anti-Rabbit IgG Secondary antibody at 1/1000 dilution for 30 minutes.Unlabelled sample was used as a control (cells without incubation with primary antibody; black).
