Recombinant COL1A1/Collagen I Monoclonal Antibody (ET1609-68)

  • Recombinant

Applications

  • WB

  • IHC-P

REACTIVITY

  • Human

  • Cow

Western blot analysis of COL1A1/Collagen Ⅰ on human placenta tissue lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody (ET1609-68, 1/500) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody (HA1001) at 1:5,000 dilution was used for 1 hour at room temperature.

  • Immunohistochemical analysis of paraffin-embedded human kidney tissue using anti-COL1A1/Collagen Ⅰ antibody. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 8.0-8.4) for 20 minutes. The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (ET1609-68, 1/50) for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.
  • Immunohistochemical analysis of paraffin-embedded human uterus tissue using anti-COL1A1/Collagen Ⅰ antibody. SEQUENCE SIMILARITIES

Belongs to the fibrillar collagen family.

TISSUE SPECIFICITY

Forms the fibrils of tendon, ligaments and bones. In bones the fibrils are mineralized with calcium hydroxyapatite.

POST-TRANSLATIONAL MODIFICATION

Contains mostly 4-hydroxyproline. Proline residues at the third position of the tripeptide repeating unit (G-X-Y) are hydroxylated in some or all of the chains.; Contains 3-hydroxyproline at a few sites. This modification occurs on the first proline residue in the sequence motif Gly-Pro-Hyp, where Hyp is 4-hydroxyproline.; Lysine residues at the third position of the tripeptide repeating unit (G-X-Y) are 5-hydroxylated in some or all of the chains.; O-glycosylated on hydroxylated lysine residues. The O-linked glycan consists of a Glc-Gal disaccharide.

SUBCELLULAR LOCATION

Secreted, extracellular space, extracellular matrix.

FUNCTION

Type I collagen is a member of group I collagen (fibrillar forming collagen).
